The innovation of this research is to combine the traditional immunohistochemical staining technique, which can accurately label multiple macromolecules in cells, with multicolor three-dimensional random optical reconstruction super-resolution microscopy (3D-dSTORM), which makes it possible to image cell wall polysaccharides with high sensitivity and resolution. dSTORM super-resolution technology, invented by Chinese scientist Xiaowei Zhuang, advances microscopy resolution to a few nanometers, which is nearly two orders of magnitude higher than the limit of optical diffraction.
